We are having problems with the peak of azithromycin, because the tail is very long and fails the tailing factor, and we could not get here in Colombia one of his preveedor columns to replace it.

Question
Could you please let us know the eluent condition for the analysis of Azithromycin?
If the eluent condition is not basic, please try the following eluent condition.
http://www.shodex.com/english/dc040244.html

And what amount of the injection volume? In this case, we think 5uL is the maximum injection volume, because the sample concentration is high.

If the eluent condition you used is the same, could you please send the chromatogram with the pressure condition and the injection volume?

Answer
1. Eluent
– 48% : It is prepared by weighing 2 g of KH2PO4 (Merck) in 700 mL
of water, mix, was added 150 ml of acetonitrile( JTBaker) and the pH
was adjusted to 11 with ammonium hydroxide.
– 46% Acetonitrile
– 6% Water
2. Detector: UV 225 nm, WV 4nm
3. Flow: 1.0 mL/min
4. injeccion volume: 25 uL
5. Oven temperature: 30?C
6. Column: Shodex ODP-50 6D (150*6)mm, 5 um

These chromatographic conditions, we obtain a pressure of approximately
1900 to 2100 psi.

->We think it is better to decrease the injection volume to 5ul or dilute the sample solution.

And the column pressure is a little too high with the eluent condition.
Approximate pressure of ODP-50 6D with this eluent condition is 600 – 700psi.
We estimate the column may have a gap at the inlet of the column.
Once this gap is made, it is impossible to repair the column.
In that case, we recommend to exchange it to the new one.
