HILIC - Polymer Based
Hydrophilic Interaction Liquid Chromatography (HILIC)
Discover the power of Hydrophilic Interaction Liquid Chromatography (HILIC) with our premium range of HILIC columns. Designed for superior separation of polar compounds, our HILIC columns offer unmatched performance and reliability for your analytical needsApplications:
- Pharmaceutical Analysis: Efficiently separate and analyze polar drug compounds and metabolites
- Biotech Research: Ideal for studying phosphorylated compounds and metabolomics
- Food & Agriculture: Quantify Organic Acids, Sugars, and Vitamins with ease
- Environmental Testing: Excellent retention of polar pesticides like glyphosate and environmental contaminates like short-chain PFAS (ex. TFA)
HILIC Fundamentals
HILIC stands for Hydrophilic Interaction liquid Chromatography. In HILIC mode, a packing material with a highly polar surface is used, with typical functional groups such as amide groups, amino groups, and diol functional groups bonded to silica or polymer base materials. Typically, a polar solvent such as a mixture of water and acetonitrile is used as the eluent, with high a organic solvent ratio to water. Under these conditions, a hydration layer forms on the packing material surface. The analyte’s distribution between the packing material and eluent is influenced by their polarities, analytes are eluted in an order of higher polarity to lower polarity. In HILIC mode, in addition to partitioning, electrostatic interactions, hydrogen bonding, and van der Waals interactions work in a complex manner to achieve high selectivity.

HILIC Series Overview
| VG-50 |
|
|---|---|
| VT-50 2D |
|
| VC-50 2D | Modified carboxyl group is suitable for cationic substance analysis including amines The dominant separation mode is RP or IEX rather than HILIC mode |
| VN-50 | The modified diol groups on the packing material create the HILIC mode Suitable for oligonucleotide and oligosaccharide separation which is not possible by SEC or conventional HILIC columns VN-50 1D adopts a double structure housing with an outer surface made of SUS and an inner surface [wetted part] made of PEEK. Covers the weak point of physical strength of PEEK housing and can be used under high pressure conditions. VN-50 1D has an inner diameter of 1.0 mm, which makes it possible to suppress the flow rate to 0.1 mL/min or less, so it suitable for high sensitivity analysis by LC/MS. |

Innovative Solutions for Your Laboratory
Shodex polymer-based columns, including ligand exchange, ion exclusion and select GFC columns, are engineered for durable performance within specified operating windows. Select polymer-based columns support wide pH operating ranges (up to pH 2–13); refer to individual column specifications for operating limits. For specific pH, solvent, temperature or counterion compatibility, always refer to the individual column datasheet. Shodex ligand exchange HPLC columns are optimized for the separation of select small, polar molecules — most commonly sugars, but also applicable to amino acids, depending on the model. The SUGAR column series is designed for high-resolution separation and quantification of mono- and disaccharides and can support oligosaccharide analysis in many food and beverage samples. For particularly complex matrices or high-oligosaccharide content, performance may depend on application-specific optimization. Shodex GFC saccharide columns separate carbohydrates according to hydrodynamic size using size exclusion mechanisms. Shodex has developed the proprietary RSpak KC-811 column, a polymer-based organic acid column that operates primarily by ion exclusion with secondary reversed-phase interactions for separation and quantification of organic acids across various matrices. Since it is a polymer-based column, it offers high chemical stability.Support for Regulated Methods
While results and lifespan can vary, Shodex columns are designed for consistent separation, supporting confidence in saccharide analysis HPLC data, as well as long-term performance. Many Shodex columns are suitable for use with organic acid ion exclusion, ligand exchange, sugar separation and regulated methods and are referenced in official compendia such as USP-NF and others. Users should verify that their chosen column is validated for the specific regulatory method required.
| Characteristics of Size Exclusion Chromatography |
|---|
| Network of pores on the surface of the packing material works as molecular sieve to separate molecules based on hydrodynamic size. |
| To separate molecules solely based on hydrodynamic size (molecular size in solution), it requires an analytical condition that prevents interactions between compounds and packing gel. |
| The bigger the molecule size, the faster the elution sequence. |
| Used for molecular weight or molecular distribution determination of macromolecules and characterization of oligomers. |
| SEC is a non-interactive separation technique; minimizing interactions between analytes and the stationary phase is critical for accurate molecular weight determination. |
